Multi-Electrode Array (MEA) system - pharmacological and functional studies of organotypic slice cultures

Fields of research:

The multi-electrode array (MEA) system is well suited for pharmacological and functional studies of organotypic slice cultures. Neuronal activities can be recorded from several different tissue areas in the same culture. The main advantage is the capacity of long-term monitoring of extracellular signals for testing of regenerative sprouting.
Currently we establish the method on organotypic hippocampal cell cultures.

Methods of research:

Organotypic hippocampal slice cultures are made according to Stoppini et al. (1991). Slices from five to ten day-old rats are cultivated for at least ten days and transferred to the multi-electrode array system. The system consists of three interconnected parts (see pictures above):
1. connecting-box (A) with biological/electronic interface (B) consisting of the perfusion chamber with multi-electrode array (C, D) on permeable membrane
2. electronic device with signal conditioner, multiplexor, and amplifiers
3. computer with data acquisition board and processing software
