Mitochondria from different tissues (brain, heart, liver) are prepared from 220-240 g male Wistar rats in ice-cold medium by using a standard procedure. Oxygen uptake of the mitochondria is measured at 30°C in a thermostat-controlled chamber equipped with a Clark-type and a TPP+-sensitive electrode. The mitochondrial membrane potential is calculated from the extramitochondrial TPP+-concentration. Respiration and membrane potential characterise the functional intactness of the mitochondria. During Hypoxia the mitochondrial membrane potential collapse partially restoring after reoxygenation indicating damage of mitochondria by hypoxia/reoxygenation. Respiration and membrane potential measurments are also used for studying ion transports through the mitochondrial membrane. Determination of the permeability of the mitochondrial membrane and measurements of enzyme activities are additional components of our spectrum of methods.
